Depth alone is an inappropriate proxy for physiological change in the mesophotic coral Agaricia lamarcki

The physiology of mesophotic Scleractinia varies with depth in response to environmental change. Previous research has documented trends in heterotrophy and photosynthesis with depth, but has not addressed between-site variation for a single species. Environmental differences between sites at a local scale and heterogeneous microhabitats, because of irradiance and food availability, are likely important factors when explaining the occurrence and physiology of Scleractinia. Here, 108 colonies of Agaricia lamarcki were sampled from two locations off the coast of Utila, Honduras, distributed evenly down the observed 50 m depth range of the species. We found that depth alone was not sufficient to fully explain physiological variation. Pulse Amplitude-Modulation fluorometry and stable isotope analyses revealed that trends in photochemical and heterotrophic activity with depth varied markedly between sites. Our isotope analyses do not support an obligate link between photosynthetic activity and heterotrophic subsidy with increasing depth. We found that A. lamarcki colonies at the bottom of the species depth range can be physiologically similar to those nearer the surface. As a potential explanation, we hypothesize sites with high topographical complexity, and therefore varied microhabitats, may provide more physiological niches distributed across a larger depth range. Varied microhabitats with depth may reduce the dominance of depth as a physiological determinant. Thus, A. lamarcki may ‘avoid’ changes in environment with depth, by instead existing in a subset of favourable niches. Our observations correlate with site-specific depth ranges, advocating for linking physiology and abiotic profiles when defining the distribution of mesophotic taxa.

Entonaema, Pulveria, Phylacia, Rhopalostroma, Sarcoxylon and Thamnomyces are relatively small and poorly studied genera of Xylariaceae. Their affinities to the mainstream of the family largely remain to be evaluated. Secondary metabolite profiles of type materials, recently collected specimens, and cultures, were generated to address this problem from a chemotaxonomic point of view. Micro-scale extraction and subsequent analytical HPLC with UV/visual (diode array) and ms detection in the positive and negative Electrospray mode were carried out, employing spectral libraries and standardised gradients that had been optimised to detect characteristic pure compounds in species of allied genera. Surprisingly, the characteristic metabolites had frequently remained stable even in specimens collected up to 190 years ago. Hence, this methodology not only proved valuable to establish the conspecificity of type materials with recent records, but also revealed some interesting correlations: (1) Stromatal pigments of Entonaema cinnabarina, E. globosum and E. liquescens are mitorubrins and other characteristic compounds also prevailing in particular species of Hypoxylon; (2) Rhopalostroma, Phylacia, Pulveria and Thamnomyces contain binapththalenes and other compounds typical of Daldinia and Hypoxylon; (3) Sarcoxylon, as well as E. dengii, E. moluccanum and E. pallida, contained none of these pigments, but characteristic yet unknown lipophilic metabolites were detected in their stromatal extracts; (4) Cultures of E. cinnabarina and Rhopalostroma indicum, obtained for the first time, produced essentially the same secondary metabolites that are also typical of Daldinia, but absent in Biscogniauxia, Hypoxylon and other xylariaceous genera. Hence, chemotaxonomic characters reflected the adaptive radiation of this family. Correlations between the evolution of morphological/anatomical characters are discussed.
Ascobotryozyma cognata sp. nov., a new ascomycetous yeast associated with nematodes from wood-boring beetle galleries

A new species of Ascobotryozyma, A. cognata sp. nov. (anamorph Botryozyma cognata), was isolated from beetle galleries in Idaho, USA. A. cognata was found on the surface of free-living nematodes, Panagrellus dubius, collected from galleries created by the long-horned beetle Saperda calcarata in Populus (aspen), and the weevil Cryptorhynchus lapathi in Salix (willow). A. cognata isolates were collected from similar habits and in relatively close proximity to those of A. americana, the only species described from North America. The recognition of A. cognata as a distinct species was supported by morphological and molecular data. Thallus cells of A. cognata were significantly shorter than those of A. americana. Low DNA reassociation values, notably different randomly amplified polymorphic DNA (RAPD), inter-sequence simple repeat (ISSR), and amplified fragment-length polymorphic (AFLP) fingerprints, and sequence divergence in both the D1/D2 domain of the nuc-LSU rDNA and an additional unidentified region were all consistent with the recognition of a new species.

In a polyphasic classification approach, stromata and cultures of Daldinia and allied Xylariaceae from around the world were studied for: (1) morphology of teleomorphs and anamorphs; (2) metabolite patterns in stromata and cultures, employing analytical HPLC-UV-visible (diode array) detection and mass spectrometric detection; (3) amplified 18S rDNA restriction patterns (ARDRA); and (4) PCR amplified minisatellite regions. Comparison of type material, herbarium specimens, cultures, and freshly collected material revealed new evidence on their geographic distribution. Several Daldinia spp. were identified in Europe and other locations for the first time. The results point towards the existence of further undescribed species. Stromata of a given species never contained the same major metabolites as corresponding cultures. Most cultures of Daldinia spp. produced naphthalene and chromane derivatives, differing from allied genera by the absence of mellein. Stromata of Daldinia spp. did not produce mitorubrin, but generally contained binaphthyls. Metabolite profiles were correlated with colours of KOH-extractable stromatal pigments. The yellow azaphilones and benzophenones found in D. childiae were lacking in species with purple stromatal pigments. Cytochalasins were found in stromata of D. eschscholzii. Genetic fingerprints helped to distinguish morphologically closely related taxa. ARDRA gave specific results for species whose 18S rDNA contained insertions, while minisatellite PCR provided specific genetic fingerprints. A combination of both PCR based techniques provided a fair resolution of genetic subtypes, reflecting the intrageneric variance in Daldinia as established from morphological data and secondary metabolite profiles.

Cultures of Penzigia cranioides and Poronia pileiformis obtained from material collected in Taiwan produced anamorphs in Xylocoremium and Lindquistia, respectively. The former is reminiscent of those produced by undoubted Xylaria species. Penzigia cranioides, the type species of the genus, is therefore accepted as X. cranioides. Penzigia thus becomes a taxonomic synonym of Xylaria. The position of P. pileiformis in Poronia is considered justified primarily on its anamorph; an epitype is designated.

Six new taxa of Biscogniauxia are described from Taiwan, including four new species: B. ambiens, B. cylindrispora, B. formosana, and B. latirima; and two new varieties: B. albosticta var. orientalis and B. formosana var. kentingensis. Their cultural and anamorphic data are also provided. A description is provided for B. doidgeae based on its type. Cultural and anamorphic data are given for B. philippinensis var. microspora. A key to the 57 known species of Biscogniauxia in the world is also given.

Nine isolates of a Colletotrichum sp. collected in North America on yellow water-lily (Nuphar luteum subsp. polysepalum) at five locations in the Pacific Northwest (PNW) and one isolate from Nymphaea odorata in Rhode Island were compared to three isolates of C. nymphaeae occurring on Nymphaea alba and Nuphar luteum in Europe. The appressoria of all North American isolates were significantly wider than those of C. nymphaeae. Conidia of the nine PNW isolates were significantly wider than those of both the Rhode Island isolate and C. nymphaeae isolates. All isolates were compared using random amplified polymorphic DNA (RAPD) and restriction fragment length polymorphism of the ITS region (RFLP-ITS). Two distinct clusters were differentiated in the dendrogram based on the RAPD analysis. The ten North American isolates formed one cluster and the European isolates of C. nymphaeae formed a second cluster. The separation of the North American isolates from the European isolates was supported by distinct restriction digest phenotypes of the ITS region. Based on morphological and molecular characterization the North American fungus is proposed as a new species, C. nupharicola.

Moelleroclavus is considered to be an anamorphic genus. The teleomorph of M. penicilliopsis is shown through field observations, collection examination, and cultures to represent a previously unnamed species of Xylaria, X. moelleroclavus sp. nov. Xylaria lingua and X. scruposa are also discussed.

Stilbohypoxylon is resurrected. The type species, S. moelleri, is redescribed and S. samuelsii is described as new. The third species, S. quisquiliarum, represents a new combination. A key to these taxa is provided.

Summary
Introduction
Modern microbial mats are structurally coherent macroscopic accumulations of microorganisms. Mats are widely distributed on earth. They are found in a surprisingly large number of diverse environments from the equatorial zones to both polar regions. They vary in size from extensive terrestrial and hypersaline mats that cover areas several square kilometers in extent to minute mats only a few square centimeters in area found in small thermal springs. They vary in thickness from massive accumulations measured in meters, such as those in the Persian Gulf and the Red Sea region, to thin films less than a few millimeters in thickness. In addition to being highly varied in size, modern microbial mats are also very diverse in morphology, community structure, and physiological characteristics. What do such mats have in common? Under what conditions do they form? What is the basis of their diversity? What insight do they provide, if any, to the interpretation of the widespread stromatolites of the Proterozoic?
A Terminology
Microbial mats are accretionary cohesive microbial communities which are often laminated and found growing at the sediment-water (occasionally sediment-air) interface. Most mats stabilize unconsolidated sediment. The mats are comprised of the various microorganisms that accumulate along with their metabolic products. The most conspicuous of these products is usually a copious amount of extracellular polysaccharide which helps hold the cells together to form a cohesive structure.
